Effects Loop vs. Line Out/In

What's the difference between an "FX" Loop and Line Out/Line In? And is it good to use them simultaneously? finally would the FX Loop be before or after the Line Out/In?

Your second question answers your first.

In general the FX loop is part of the preamp stage and as such your pre out will include all your FX loop components.

Some amps actually turn off power to the preamp section when there is an input in the power amp in section, but you can easily check if this is the case with your amp, by placing an input into the preamp and a patch between the pre out and power in.
